This kit provides a rapid, sensitive, and economical tool to simultaneously detect the relative levels of 53 angiogenesis-related proteins in a single sample. Analytes include soluble growth and differentiation factors, extracellular matrix components, proteases, membrane-bound receptors, and intracellular signaling molecules. The use of R&D Systems antibody arrays requires no specialized equipment and eliminates the need to perform multiple immunoprecipitation/Western blot experiments.

General Assay Principle
Carefully selected capture antibodies have been spotted in duplicate on nitrocellulose membranes. Cell culture supernates, cell lysates, tissue lysates, or serum samples are diluted and mixed with a cocktail of biotinylated detection antibodies. The sample/antibody mixture is then incubated with the array. Any protein/detection antibody complex present is bound by its cognate immobilized capture antibody on the membrane. Streptavidin-Horseradish Peroxidase and chemiluminescent detection reagents are added, and a signal is produced in proportion to the amount of cytokine bound. Chemiluminescence is detected in the same manner as a Western blot.

| Multiple Proteins in TNF-alpha-treated Balb/3T3 Cells were Assessed using the Mouse Angiogenesis Array. A. The Proteome Profiler Mouse Angiogenesis Array Kit (Catalog # ARY015) was used to simultaneously assess the relative levels of 53 mouse angiogenesis-related proteins in cell lysates from Balb/3T3 mouse embryonic fibroblast cells that had been left untreated or treated with recombinant mouse TNF-alpha (Catalog # 410-MT) for 24 hours. B. Histogram profiles for select proteins were generated by quantifying the mean spot pixel density from the arrays using image software analysis.
